Bone Tissue Engineering Utilizing Adult Stem Cells in Biologically Functionalized Hydrogels

چکیده
The goal of these studies was to investigate the effect of resveratrol treatment on the osteogenic potential of human and rat adipose derived stem cells both in a 2-D and 3D culture environment. Adipose derived stem cells (ADSCs) have shown promise as a potential source of osteogenic progenitor cells. Resveratrol has been shown to inhibit adipogenic differentiation while simultaneously activating osteogenic differentiation. 2D experiments with rat ADSCs identified 25 μM of resveratrol as an effective dose to increase the production of osteogenic genes. The effect of 25 μM resveratrol treatment was investigated in a pre-treatment and continuous treatment regimen in 3-D culture and demonstrated that pre-treatment was sufficient to increase the mineralized matrix production of rat ADSCs. We next sought to investigate whether this result was also observable with human ADSCs and found that the human cells did not respond to 25 μM

Bone Tissue Engineering: a Mini-Review
Despite advances in bone tissue engineering, auto grafts from intra-oral or extra-oral donor sites are still the gold standard for treatment of large craniomaxillofacial defects. Biomaterial development, application of growth factor, and stem cells, open new gateway to bone regeneration studies, but real translation from bench to bedside have not yet happened. In this review article, a number o...
متن کاملAdult Mesenchymal Stem Cells for Tissue Engineering
The identification of multipotential mesenchymal stem cells (MSCs) derived from adult human tissues such as bone marrow and connective tissues has provided exciting prospects for cell-based tissue engineering and regeneration. This review article focuses on the biology of MSCs, their differentiation potentials in vitro and in vivo, and their application in tissue engineering...
متن کاملEngineering cartilage or endochondral bone: a comparison of different naturally derived hydrogels.
Cartilaginous tissues engineered using mesenchymal stem cells (MSCs) have been shown to generate bone in vivo by executing an endochondral programme. This may hinder the use of MSCs for articular cartilage regeneration, but opens the possibility of using engineered cartilaginous tissues for large bone defect repair. Hydrogels may be an attractive tool in the scaling-up of such tissue engineered...
متن کاملReview Paper: Embryonic Stem Cell and Osteogenic Differentiation
Bone tissue engineering has been one of the most promising areas of research, providing a potential clinical application to cure bone defects. Recently, various stem cells, including embryonic stem cells (ESCs), bone marrow-derived mesenchymal stem cells (BM-MSCs), umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs), adipose tissue-derived stem cells (ADSCs), muscle-derived stem cell...
متن کاملFunctionalized α-Helical Peptide Hydrogels for Neural Tissue Engineering
Trauma to the central and peripheral nervous systems often lead to serious morbidity. Current surgical methods for repairing or replacing such damage have limitations. Tissue engineering offers a potential alternative. Here we show that functionalized α-helical-peptide hydrogels can be used to induce attachment, migration, proliferation and differentiation of murine embryonic neural stem cells ...
